{"entity":{"id":"docetaxel","kind":"drug","name":"Docetaxel","aka":[],"tldr":"The first chemotherapy to extend life in prostate cancer (2004), now part of triplet therapy at first metastatic diagnosis.","summary":"Docetaxel is a taxane that stabilises microtubules and arrests dividing cells; in prostate cancer it also disrupts androgen receptor nuclear trafficking. It was the first chemotherapy to extend life in metastatic castration-resistant prostate cancer (TAX 327, 2004) and later moved to first metastatic diagnosis: CHAARTED (OS 57.6 versus 44.0 months, HR 0.61) and STAMPEDE showed an overall survival benefit in high-volume hormone-sensitive disease, and PEACE-1 and ARASENS built triplets by adding abiraterone or darolutamide. It is also widely used in breast, lung, gastric and head and neck cancers. Neutropenia, neuropathy and fluid retention limit its use in frail patients, and whether men with low-volume metastatic disease gain anything from it remains contested. For a newcomer, it is the workhorse chemotherapy given every three weeks across many cancers.","status":"approved","asOf":"2026-09-06","wikipedia":"https://en.wikipedia.org/wiki/Docetaxel","links":[{"label":"FDA label (DailyMed)","url":"https://dailymed.nlm.nih.gov/dailymed/search.cfm?query=Docetaxel"}],"tags":[],"related":[],"indications":["prostate","prostate-mhspc","prostate-mcrpc"],"sections":[],"technologies":[],"targets":[],"drugs":[],"companies":[],"institutions":[],"pathways":[],"terms":[],"trials":["chaarted","stampede","nct06520345","nct07590934"],"people":[],"bottlenecks":[],"keyPapers":[],"journals":[],"dependsOn":[],"notes":[],"brand":"Taxotere (generic)","modality":"Cytotoxic chemotherapy (taxane)","mechanism":"Microtubule stabilisation; also disrupts AR nuclear trafficking.","approvals":[{"region":"US","year":1996,"indication":"Locally advanced or metastatic breast cancer after chemotherapy (Taxotere, NDA 020449)"},{"region":"US","year":2004,"indication":"Metastatic CRPC (with prednisone)"}],"mechanismSteps":[],"toxicity":[],"access":[],"regulatoryEvents":[]},"route":"/drugs/docetaxel/","neighbours":{"indication":[{"id":"prostate-mcrpc","kind":"indication","name":"Metastatic castration-resistant prostate cancer","route":"/indications/prostate-mcrpc/"},{"id":"prostate-mhspc","kind":"indication","name":"Metastatic hormone-sensitive prostate cancer","route":"/indications/prostate-mhspc/"},{"id":"prostate-nepc","kind":"indication","name":"Neuroendocrine and small-cell prostate cancer","route":"/indications/prostate-nepc/"},{"id":"osteosarcoma","kind":"indication","name":"Osteosarcoma","route":"/indications/osteosarcoma/"},{"id":"prostate","kind":"indication","name":"Prostate cancer","route":"/indications/prostate/"}],"trial":[{"id":"chaarted","kind":"trial","name":"CHAARTED (E3805)","route":"/trials/chaarted/"},{"id":"nct07590934","kind":"trial","name":"Phase Ib/II Platform Study of Multiple Anti-Cancer Agents in Participants With Metastatic Prostate Cancer","route":"/trials/nct07590934/"},{"id":"stampede","kind":"trial","name":"STAMPEDE","route":"/trials/stampede/"},{"id":"nct06520345","kind":"trial","name":"The Study of 177Lu-TLX591 Plus SOC Versus SOC Alone in Patients With mCRPC (ProstACT Global)","route":"/trials/nct06520345/"},{"id":"upfront-psma","kind":"trial","name":"UpFrontPSMA","route":"/trials/upfront-psma/"}],"pairing":[{"id":"docetaxel-then-lu177-psma","kind":"pairing","name":"Docetaxel (taxane chemotherapy) then 177Lu-PSMA-617","route":"/pairings/docetaxel-then-lu177-psma/"}]}}
